摘要:桑樹桑黃是一種具有良好藥效的藥用多孔菌,其多糖是主要活性成分之一,具有抗腫瘤、抗氧化、免疫調(diào)節(jié)和其他藥理作用。為提高桑樹桑黃菌絲體干重及桑黃多糖得率,采用單因素和正交試驗對影響桑黃菌絲體干重的培養(yǎng)基配方及液體發(fā)酵培養(yǎng)條件進行優(yōu)化;利用超聲波輔助提取法對桑黃多糖提取工藝條件進行優(yōu)化。結(jié)果表明,桑樹桑黃最佳培養(yǎng)基配方為馬鈴薯(去皮)200 g·L-1、乳糖32 g·L-1,胰蛋白胨10 g·L-1,NaCl 2.00 g·L-1,MgSO4·7H2O 1 g·L-1,VB1 10.0 mg·L-1,獲得菌絲體干重為2.82 g·L-1。在溫度28 ℃,接種量9%,轉(zhuǎn)速180 r·min-1,pH6.0條件下,最大菌絲量為3.12 g·L-1。桑黃多糖最佳提取工藝為超聲波功率200 W,料液比1∶60 g·mL-1,提取時間40 min,溫度50 ℃,此時桑黃多糖得率為5.54%。說明本研究優(yōu)化的桑樹桑黃培養(yǎng)基配方、發(fā)酵條件及多糖提取工藝合理、可行。

Abstract:Sanghuangporus sanghuang is a medicinal porous bacterium with good medicinal efficacy, and its polysaccharide is one of the main active ingredients, with antitumor, antioxidant, immunomodulatory and other pharmacological effects. In order to improve the dry weight of S. sanghuang mycelia and the yield of S.sanghuang polysaccharide, single factor and orthogonal test were used to optimize the medium formula and liquid fermentation culture conditions affecting the dry weight of S.sanghuang mycelia. Ultrasonic assisted extraction method was used to optimize the extraction conditions of polysaccharide yield of S.sanghuang. The results showed that the optimal medium formula of S. sanghuang was potatoes (peeled) 200 g·L-1,lactose 32 g·L-1, tryptone 10 g·L-1, NaCl 2.00 g·L-1, MgSO4·7H2O 1 g·L-1, VB1 10.0 mg·L-1, and mycelium dry weight was 2.82 g·L-1. Under the conditions of temperature 28 ℃, inoculation amount 9%, rotational speed 180 r·min-1 and pH6, the maximum mycelium amount was 3.12 g·L-1. The optimal extraction process was as follows: ultrasonic power 200 W, solid-liquid ratio 1∶60, extraction time 40 min, temperature 50 ℃, the yield of polysaccharides was 5.54%. The results showed that the optimized culture medium formula, fermentation conditions and polysaccharide extraction technology of S. sanghuang were reasonable and feasible, which could provide reference for liquid fermentation of S. sanghuang mycelium to produce polysaccharide.
